چکیده

To elucidate the effects of microRNA (miRNA) regulation in skeletal muscle of adult pigs, miRNA expression profiling was performed with RNA extracted from longissimus dorsi (LD) muscle samples from 174 F2 pigs (~ 5.5 months of age) from a Duroc × Pietrain resource population. Total RNA was extracted from LD samples, and libraries were sequenced on an Illumina HiSeq 2500 platform in 1 × 50 bp format. After processing, 232,826,977 total reads were aligned to the Sus scrofa reference genome (v10.2.79), with 74.8% of total reads mapping successfully. The miRDeep2 software package was utilized to quantify annotated Sus scrofa mature miRNAs from miRBase (Release 21) and to predict candidate novel miRNA precursors. Among the retained 295 normalized mature miRNA expression profiles ssc-miR-1, ssc-miR-133a-3p, ssc-miR-378, ssc-miR-206, and ssc-miR-10b were the most abundant, all of which have previously been shown to be expressed in pig skeletal muscle. Additionally, 27 unique candidate novel miRNA precursors were identified exhibiting homologous sequence to annotated human miRNAs. The composition of classes of small RNA present in this dataset was also characterized; while the majority of unique expressed sequence tags were not annotated in any of the queried databases, the most abundantly expressed class of small RNA in this dataset was miRNAs. This data provides a resource to evaluate miRNA regulation of gene expression and effects on complex trait phenotypes in adult pig skeletal muscle. The raw sequencing data were deposited in the Sequence Read Archive, BioProject PRJNA363073.

Combining QTL- and expression-analysis: identification of functional positional candidate genes for meat quality and carcass traits

In order to identify genes controlling meat quality and carcass traits we are aiming to combine two approaches: (1) the QTL analysis (2) and expression profiling. Here results of QTL analyses in different porcine resource populations and current attempts to identify functional candidate genes for meat quality traits are reviewed and own efforts are presented: A QTL scan for meat quality and car...

Content of lipid components in m. longissimus dorsi of progeny of the boars descending from reciprocal crossing of the Pietrain and Duroc breeds

The study has been carried out on 72 fatteners being the offspring of crossbreed sows x Pietrain boars (group I) and hybrid boars of the ♂ Pietrain x ♀ Duroc breeds (group II) and, on the ♀ Duroc x ♂ Pietrain breed (group III). In m. longissimus dorsi, the content of dry matter, protein, intramuscular fat and total cholesterol has been determined. In the intramuscular fat of the muscle, the com...

Association of Insulin-like growth factor binding protein 2 genotypes with growth, carcass and meat quality traits in pigs

BACKGROUND This study was conducted to investigate the potential association of variation in the insulin-like growth factor binding protein 2 (IGFBP2) gene with growth, carcass and meat quality traits in pigs. IGFBP2 is a member of the insulin-like growth factor binding protein family that is involved in regulating growth, and it maps to a region of pig chromosome 15 containing significant quan...

Identification of quantitative trait transcripts for growth traits in the large scales of liver and muscle samples.

Growth-related traits are economically important traits to the pig industry. Identification of causative gene and mutation responsible for growth-related QTL will facilitate the improvement of pig growth through marker-assisted selection. In this study, we applied whole genome gene expression and quantitative trait transcript (QTT) analyses in 497 liver and 586 longissimus dorsi muscle samples ...

Gene expression profile of Musculus longissimus dorsi in bulls of a Charolais × Holstein F2-cross with divergent intramuscular fat content

Transcriptomes of Musculus longissimus dorsi (MLD) were compared between bulls from a F2-cross derived from Charolais and Holstein Friesian. Two groups of 10 bulls were selected which differed significantly in intramuscular fat (IMF) deposition despite standardized husbandry and feeding conditions and identical sires in both groups. Consequently, genetic factors underlying the different capabil...
